FDA-approved September 2014. REWIND (N=9,901) demonstrated CV benefit including primary prevention. AWARD program showed A1C reductions of 0.8–1.6%. Weight loss ~3–5 kg, modest vs. semaglutide.

§ 02   Mechanism

Mechanism of action

Recombinant GLP-1 analog fused to a modified human IgG4-Fc domain via a small peptide linker. The Fc fusion extends half-life to ~5 days, enabling once-weekly dosing. Activates GLP-1R to enhance glucose-dependent insulin secretion, suppress glucagon, delay gastric emptying, and promote satiety.
