Overview
Synthetic gonadotropin-releasing hormone (decapeptide). Historically FDA-approved for HPG-axis testing and pulsatile ovulation induction. Today compounded off-label as an HCG alternative in TRT / post-cycle protocols.
Factrel (NDA 018123) approved 1978 for diagnostic HPG testing; marketing discontinued. Lutrepulse approved for pulsatile ovulation induction; no longer marketed. Currently compounded.
§ 02 Mechanism
Mechanism of action
Gonadorelin is identical to native GnRH. Pulsatile administration (every 90–120 min) activates pituitary GnRH receptors to release LH and FSH, driving endogenous testosterone or ovulation. Continuous dosing paradoxically downregulates GnRH receptors (the principle behind GnRH-agonist cancer therapy).
